What is the best method to isolate maximum number of cells from whole blood besides using ficoll hypaque for lymphocyte cultures. Also please share the experience of optimum concentration and duration of PHA and ConA for cell stimulation.
For lymphocyte proliferation and functional assays, peripheral blood, obtained in an acid citrate dextrose (ACD, yellow top) tube, is the best to use. Heparinized blood (green top tubes) may be used, but in general, PBMCs prepared from heparinized blood tend to clump and may incorporate slightly less H3-thymidine than cells from ACD- anti-coagulated blood.
