By UV-vis: materials with a direct band gap, there will be a sharp increase in absorption at the energy corresponding to the band gap. In contrast, for materials with an indirect band gap, there will be a gradual increase in absorption as the energy approaches the band gap, followed by a sharp increase in absorption at a higher energy level.
By PL spectroscopy: For materials with a direct band gap, the emitted light will have a energy level corresponding to the band gap. In contrast, for materials with an indirect band gap, the emitted light will have a lower energy level than the band gap.
