I grow HeLa in Advanced MEM, After 1-2 days I see black bodies inside the cells as well as outside the cells. I checked at 40x and doesn't look like tumbling/ bacterial contamination. can any one explain? I want to use it for nuclear extract preparation.
I guess they may be either mycoplasma and some debris from medium or serum.
Yes, it will be cell debris. You aren´t able to see a mycoplasma contamination. Because of the fact, that the HeLa cells are tumor cells, they are growing very fast. You should split them before they reach 100 % confluency. If the cells grow in a normal manner, you don´t have to be afraid. After two days of cultivation, you should see a bacterial contamination (except a mycoplasma contamination). Are the black bodies moving or are they stationary?
It might be due to over-confluent monolayer of HeLa cells. Maybe you should try to lower your seeding density.
