I generated the MDA MB 231 stable expressing HER2 mutations with MSCV and HER2 WT as negative and positive control, respectively. Then, I injected 1x10^4 cell of each clone to SCID mice. Unfortunately, HER2 WT formed the smallest tumor while vector control and mutations showed more bigger. The same result also got when doing in nude mice.
In vitro, my HER2 mutations showed the lower migration, invasion, spheroid formation ability in comparison with HER2 WT
So, MDA MB 231 isn't suitable for my model?
MDA MB 231 is triple negative breast cancer cell line ( Estrogen, HER-2 and Androgen receptors are absent) so I think MDA MB 231 is not a suitable model.
If your intention is to study the role of HER2, you should use triple negative breast cancer cells and overexpress HER2-WT or mutant gene. I think you have chosen correct cells but to confirm your results you should use another similar cell lines like MDA-MB-468 and DU4475.
Yes, I want to study the role of HER2 mutant. But the problem is I cannot get the consistent results between in vitro and in vivo. HER2-WT overexpression does not show the highest ability to form the tumor as itself should have.
I got 4 clones of stable expressing HER2-WT, but between themself also had the difference. The clone showed the higher sphere formation, had the lower migration & invasion ability, and vice versa
