I would like to take temporal samples of DNA from a population over time to determine whether a decline in various measures of genetic diversity is occurring. For example, samples taken at time equals 0, 5, 10, 15, and 20, or perhaps just taken at 0 and 20. For most statistical tests (for example mann whitney test) it is my understanding that samples should be independent, so, are these samples considered independent? What statistical test would be most appropriate for analyzing this data? Do you know of papers that have done so? I do not aim to use advanced coalescent methods such as BEAST, but rather want a simple statistical comparison (I will be simulating many datasets). Assume multiple loci are typed, for multiple individuals. Thank you.
