Thanks for the info. One of heterochromatic sequences is tandemly repeated 45S rDNA, which is about 45 kb each repeated unit in mammals. High sequencing accuracy is required to assemble the repeats in right order. I have not yet seen any demonstration of such effort from PacBio. My understaning is that PacBio is also exploring a nanopore technology other than enzyme-based SMART platform. Could it be that the enzyme-based sequencing technology has reached its limit?

Thank you for the attached article and book. They are excellent reviews for short-read sequencing technology. I am particularly interested in a technology which can be advanced to sequence long repetitive elements. Gross changes of these genome regions have been linked to evolution, development, physical and mental diseases, and adaptation. I see some potential for solid-phase nanopore and electron microscope sequencing technologies. I welcome any suggestion.
