I'm growing Caco-2 cells on microporous membranes, 12-well transwells, 12mm inserts, 0.4micrometer pore size. Does anyone know what the TEER value should be for fully differentiated Caco-2 cells after 21 days in culture? I'm measuring the resistance with a Millicell ERS from Millipore.

Caco-2 cell line is heterogenous and properties can vary from lab to lab. Culture conditions can also have a substantial effect. In our hands Caco2 cells grown on Costar Transwell inserts (polycarbonate, 12 mm diameter, 0.4 microm pore size) have a TEER of 600-1000 Ohmx cm2 by 14-21 days of culture. If cells are selected with vinblastin TEER can double, and easily reach 2000 Ohmxcm2. Please see our related paper (PMID: 22906709). Your results seem to fit with the VB-selected subline of Caco-2, so it is not so unusual.
To be sure that your measurement is correct, please check (i) the batteries of Millicell ERS (ii) if your electrode has been properly maintained and lightly sanded with the provided sandpaper, (iii) the built in resistance (should be 1 kOhm). If everything is OK, then your result is correct, and your lab has a Caco-2 subline with high resistance.

first of all: I haven't worked with Caco yet but I would recommend the following proceeding to investigate what resistance you are aiming for:
First I would consult the original publication of the cell line which in this case is the work by Hidalgo et al. (PMID: 2914637). I couldn't find the PDF since Gastroenterology seems to miss the link. Other publications from his group might also hold the standard values.
Secondly it seems to me that your resistance/confluence would depend on the culture protocol. I would look into the publications which established the protocol you are using.
And last but not least from a quick search in Google books I found a rough reference to a resistance around "250-400/cm2" for Caco-2 in "Epithelial Cell Culture Protocols" by Clare Wise. I don't know for which culturing technique, what permeable support and what day of growth though.

Thank you! My cells have a TEER value of 2500 ohms/cm2 and that's what's confusing me. I've seen so many different values in articles so i don't know which value to compare with. The growth area I use is about 1cm2so that value won't affect the TEER unit when I calculate according to the manual of the Millicell ERS,
TEER=(R-Rblank)*growth Area.
