Most studies in literature using HT22 cells and excitation using glutamate are viability studies. I want to stimulate these hippocampal like neurons but not induce cell death.
Please study two articles in the attachment to my letter. In one article, the question of the side effects of glutamate in very small doses in PC12 cell culture is analyzed. A similar analysis is presented in the second article on HT22 cells.
Thank you for those articles. Low doses like 10uM glutamate (used in the first paper) was what I wanted to see in HT22 cells, where I have not seen any papers use concentations less than 1mM. Do you have any idea if HT22 cells are more sensitive to glutamate concentrations than PC12 cells? This will help me choose doses to make a dose response curve for my experiments...
Dear Malavika! You are absolutely right. Linear cell cultures have often different sensitivity to damaging factors. Therefore, your opinion on the decrease of the molarity of the glutamate solution is quite true. You will detail in the methodology of the article all this, and the reviewers will understand and support the logic of your reasoning. And most importantly, you do not have to take measures to reduce the toxicity of glutamate. This will distort the results of your interesting experiments. All the best.
