Can I put an insert size of 5.7kb into a vector which has a cloning capacity of 5.4kb. This vector is a retroviral vector system.
I want to know if this is possible and what does the cloning capacity indicate? If I try then would I loose any significant region of my insert or will it be difficult to generate the virus particle with my gene of interest ?
In viral systems the capacity usually refer to the maximum length that can be packaged into virions so if you exceed this you will significantly reduce your virus yields.
You can clone it into the viral vector easily enough and make a plasmid. However, as stated above, the cloning capacity for viral vectors refers to the maximum size of the viral genome that can be packaged into infectious virions. You are only slightly over, so you may still get virions but the titer of the virus may be lower than typically for this viral strain.
