My target gene was sequenced and the sequence was correct. WB, qp and IF experiments all proved that it was overexpressed. However, when I looked for the gene regulated by it, I found no difference between it and the control. Even if I verify some published articles, I can't verify the corresponding results. I wonder why? Is there a solution?Only its own target gene was changed, and other regulated target genes were no different from the control. I also tried various transfection times, but none of them were solved.I couldn't make any difference between the proteins in the classical signaling pathways that were common in the lab and the genes that might be regulated by them in the database. Except that my target gene is expressed enough. I don't know what the problem is, and I have no way to fix it. Does anyone know how to handle this situation?
