Both of the SV40 Poly As are in the same direction and about 50 bp apart. When I cloned my donor plasmid I didn't realize there was a random sequence already there. Should I expect a significant decrease in the expression of the protein as a result of this? I've already created an edited cell line with this gene so fixing it now is not a great option.
