Our lab is going to use ISO 12966-2 and 12966-4 and there is one moment in sample preparation (rapid transmethylation method under alkali-catalysed conditions) that I can't understand. It is about the addition of saturated sodium chloride solution after transmethylation reaction. Maybe I'm not so good in chemistry to understand the purpose of this action. So, can anyone please help me to get it?

If I understand correctly, the detection technique of volatiles according to the protocol is done using headspace gas chromatography. In that case, addition of salt helps partitioning of volatiles from the matrix into headspace.

No, we don't use headspace for this analysis. Just injection of liquid sample (1 mcl) of FAMES in isooctane. So, i guess it is not that case. However, I think you might be right, but volatiles would rather move to isooctane layer than stay in glycerol.
