Dear Sr Luis, Thank you! We diagnose SBP according to this criteria also. But allow specify. What method do you use for counting of PMN in ascitic fluid? It's the manual counting in microscopy or an automated cell counter?
Dear Dr. Michiya, thank you. For diagnostic of SBP we used counter NIHON KOHDEN 6410К. Unfortunately, this counter sometimes didn’t differentiate leukocytes (in samples with low level of total cells). We compared the determination of PMN count in the ascitic fluid obtained by the manual and the automated methods. We have found out one false negative result. Is validation required for other automated blood counters? How often have you met false negative results at the relatively low levels of PMN in ascitic fluid? Is the automated blood counter reliable method for diagnostics SBP?
Dear Dr. Gavrilenko, Thank you for your reply. As far as my experience, automated cell counter did not show false-negative results. We routinely use this for the diagnosis of peritoneal dialysis related peritonitis. The sensitivity of this automated machine is quite high and it could count less than 100 per microliter cells in a ascitic fluid.
I think manual cell counter is a best method. I was see sometimes strong difference betwen this 2 methods and in my experience normally the manual method y more consistent with the others clinical and laboratories parameters
