I'm trying to revive U937 cells, but I'm having a problem getting the cell line to develop normally.
The growth media is RPMI-1640 from Sigma Aldrich with L-glutamine, 10% FBS added, as well as antibiotics, pH (7.2-7.4).
The cell line from cryopreservation was previously done in the lab, (preserved in liquid nitrogen) about 6 months ago. However, I am trying to revive the cells and culture again from different passage numbers, unfortunately, they look healthy and good for the first 24 hours but then shriveled significantly and die.
I had checked for mycoplasma contamination and it was clear.
what other conditions other than the nutrients and mycoplasma shall I check? and what is the optimum PH for U937?
