fastest can be Exoquick and you dont need ultra centrifuge. However you need to think about what you going to do with these exosomes. If it is for functional test, probably ultracentrifugation might be the best...
The gold standard method is Ultracentrifugation, but it is absolutely time-consuming and needs the expert operator and some apparatuses. There are some commercial kits, which are very simple, ready-to-use and time-saver. These days Size Exclusion Chromatography (SEC) based procedures and kits are really popular. Some Kits are used SEC-based columns to better purification (e.g, qEV (iZON), Exo-Spin (Cell Guidance Systems, etc.)). You can use SEC as a reliable and reproducible method in exosome isolation. Good Luck
Here I recommend Creative-Biostructure’s exosome engineering platform. They have developed five groups of exosome isolation techniques, including ultracentrifugation-based techniques, size-based techniques, immunoaffinity capture-based techniques, exosome precipitation, and microfluidics-based techniques. I feel it is pretty good, you can try.
