What causes micro plate reader (soft max pro) growth experiment using presto blue to fail?

under microscope, there was difference between treated cells with cytotoxic drug and control in growth and number but when I did growth and proliferation assays numbers do not make sense ( absorbance of control vs treated). Setting was correct. Protocol growth and proliferation Glo . Display reduced . What can cause this experiment to mess up? . Presto blue also was correct. 1:9 dilution with medium.
