I am looking for a non–fluorescent mutant of GFP, which should be mutant folds to a stable, native‐like structure but lacks fluorescence, should be able to tag with protein and express purify without any aggregates or folding issues like wtGFP. Is there any available on Addgene?
I am not sure if this approach would suit your particular application, but using the large fraction of the Split-GFP tag might work for you. The Large GFP(1-10) is non-fluorescent. It is commonly used to study two protein's interaction (or aggregative prone proteins) in vitro, by co-expressing another protein of interest tagged with the other small GFP fragment GFP(11), which then becomes fluorescent when they bind. It is available on Addgene in many varieties (pcDNA3.1-GFP(1-10), (Plasmid #70219)). I am not sure, but i think the large GFP(1-10) can be immunoprecipitated with certain antibodies (but you would have to confirm this). Hope i could help. Good luck.
