Perhaps it is a problem related to an incorrect pH due to the time medium has been used, its storage conditions or due to the preparation of the medium or its additives, another possibility to take into account would be to check the conditions of the CO2 incubator.
Thanks for your comment. I am collecting exosome from mdamb231’s culture medium (DMEM+1%BSA+Antibiotic). I am not familiar with crystals. What exactly are they? Like salt crystal? They do not dissolve after resuspend them in pbs.
I am not very familiar with crystals too, but it might be crystals formed from PBS. So, did you wash exosomes with PBS or Saline? Alternatively, it can be deposits of BSA or antibiotics. My suggestion is to remove the BSA and antibiotics, and use serum and antibiotic -free medium to secrete the exosomes.
PBS contains phosphate and bivalent metals like Ca and Mg, which are prone to precipitation. They could be detected in the crystal by standard procedures of analytical inorganic chemistry.
