Hi all,
I'm aiming to study a bacterial purified protein known to be toxic in the periplasm.
I haven't been able to predict the antibacterial activity of my protein by computational analysis and I would try to test it in different toxicity assays.
I thought to start with a lysozyme activity assay.
Do you know any other test I could use?
Any other toxic activity located in the periplasm?

With antibody scFv fragments, I found that both those that fold well (close to 100% soluble expression) and those that fold so poorly that they immediately formed periplasmic inclusion bodies (close to 100% insoluble) are not toxic. The problematic ones were the intermediate ones, presumably the ones that hung around in a partly folded state and potentially formed aberrant disulphide bonds to other proteins. (smear on non-reducing PAGE).
