Crab tree positive effect of Ecoli leads to acetate accumulation. Apart from engineering of Ecoli strains, what are the feeding strategies and process control optimization can be done to avoid acetate (
The best solution is controlling feed based on dissolved oxygen. Keep dO2 at >90% and glucose at less than 1 g/l. You could also try growing it on a different energy source such as glycerol.
I think acetate accumulation doesn't depend on oxygen consumption rate. I guess glucose monitoring within critical limit will reduce the acetate. Any feeding strategies and novel process conditions are available to reduce the acetate accumulation?
You can control your process by using defined media with glucose as carbon source. Employing fed-batch process (limiting glucose feeding) for controllling u (mu) below 0.3 h-1 in a pseudo-steady state and keeping glucose concentration in zero (0), you can control acetate production. Try not to limitate dO2 using enriched oxygen air (20-80 %) and use cascade O2 control on a bioreactor.
Mr Phillip Brumm if you know the correct answer you answer to the question,as per your reply to this question I can quess that you dont have minimum knowledge about fermentation process. Can anybody in the world maintain DO above 90% and that too maintaining DO is no way related to acetate production please mind it.
