What are the possible interactions and results I can look for while performing T-cell/cancer cell co culture studies?

I work in cancer biology and wish to do onco-immunology studies. I want to check weather knocking down my gene of interest has any effect on Tcell immunity in vitro. Our lab has still not optimised 3D cell culture so I want to do these studies in 2D monolayer culture.

Can anyone Suggest what possible things I can look for and how
