Hi Ofir,

I just briefly wanted to email concerning the characterization of the AIRE knock out. If it is of any help, I would recommend to follow the attached paper from Anderson et al.. They did a homemade ELISA (for the protocol see the attached methods) for circulating anti-thyroglobulin autoantibodies (with bovine thyroglobulin as antigen) and checked cellular infiltrates into the target organ, while at the same time staining for autoantibodies in tissue sections. The only thing you needed to procure is serum from MRL.lpr/lpr mice to have a good positive control. I hope that helps and good luck with your experiment.
