Hi,

I was performed TCID50 for influenza infection. It seems to me that the infection medium results in cytopathic effects, so I can't count the positive and the negative wells.

The infection medium contain:

DMEM high glucose 4.5g/L (Gibco)

0.2% BSA

25mM HEPES

1ug/ml TPCK (Sigma)

Antibiotics

However, the cells grow abnormally even without the TPCK presence.

Attached, please find 4 pictures from the same 96 well 3 days after changing the medium:

a) Normal growth medium (10% FBS)

b) Infection medium without TPCK

c) Infection medium with TPCK and 1:100 Virus (PR8)

d) Infection medium with TPCK
