I am currently differentiating PBMC (Monocyte) to iDC using GMCSF and IL4 for 5 days with the change of media on day three by substituting half of the media with fresh media containing cytokines. On day 5 I see some cells (DCs) attached to the plate and some floating. The markers I have for Monocyte and DC are HLA-DR, CD14, CD80, CD83, and CD86.

-What is the difference between attached and floating cells?

-Can they both be used together for experiments?

- What are the expression levels of these markers in Monocyte, immature DC, and mature DC?
