I am working on generating pericytes and SMCs but since both have common origin it is difficult to distinguish between the two lineage best on markers. Does anyone know of very specific markers for each of the lineage?
Pericytes are specialized mesodermal cells which line the blood vessels. Besides the early mesoderm marker CD31 for pericytes, there is a helpful chart in the millipore site below that will show you the different markers that will help you identify the more mature smooth muscle cells may be using Desmin. I would suggest you may also check out this histology site which may of help to identify the cell morphology and location. Also, try the BD Biosciences site for selection of these markers. Hope these will be of help to you.
I would go for double staining: NG2 with DAB + CD31 with permanent red. Then you see pericytes by NG2 and confirm them by morphology by seing blood vassels stained by CD31.

I would like to share this paper with the histologic, immunohistochemical, and ultrastructural features of 12 canine perivascular wall tumors of uncertain classification are boarded.
I am always trying to find a stable marker for pericytes in the BBB. Joan Abbott and others have published several in this area. You may want to look at her work. Sen gave you an excellent paper to start.
