To gather the spores, pick a frond or portion of a frond and place it between two sheets of white paper. If ripe, the spores should drop within 24 hours and will leave a pattern on the paper. Or place a frond in a paper bag. Over the course of the next few days, the spores should fall from the frond and collect in the bottom of the bag :)
In my opinion, there are two questions, one how to obtain spores from a plant? another as spore density calculated in culture?
If so: fertile pinnae are collected with closed sporangia and placed in an absorbent paper envelope, left to dry at room temperature and then the content is passed through a 0.74 mm sieve to obtain clean spores.
for the second question: spores are sown in an agar plate, dispersed with a fine brush and then five points are taken at random (1 cm2), the count is made, the summation is made and the average is taken out.
