I'm going to extract RNA from cell culture and I have to put RNA-containing microtubes in the water bath for 10 minutes at 60°C. I want to ask if any Rnase contamination danger threaten the RNA or not.
Well, the short answer is RNase presence is always a possibility. You'll want to avoid getting any water in your tubes. You can't keep the water bath itself sterile and RNase free. But, now might be a good time to clean out the water bath and start with nice clean water.
If you have heat block that can be a good alternative heat source (still not guaranteed to be RNase free but at least you can't get water leaking in).
More likely sources of RNase are you (your hands) and the micropipettes.
