I use a Sony SH 800 sorter to sort differentiated tumor cells (CD44-CD24+) of the T47D line. According to our and literature data, the CD44-CD24+ population in the T47D line is 70-80%, however, after sorting, we always get very few CD44-CD24+ cells. In order to isolate 1,000,000 CD44-CD24+ cells, we have to take 40,000,000 T47D cells. This also happens when working with other cell lines, the cell yield is always very low. I use pressure 5-6. Has anyone faced this problem? Can anyone give advice?
