Our hybridomas are facing a contamination issue.
We use RPMI1640 (Corning), 20% FBS (Cytiva), 1% PS (Gibco), and 1% ITS (Gibco) for hybrid cell culture.
After 14 days of culture, we observed mobile and swimming organisms in 96-well plate cell cultures under the microscope (Attached video).
- Are we facing bacteria in our hybridomas cell culture?
- We're considering using Normocure (Invivogen, cat. ant-noc) for Microbial contamination elimination. Did anyone try Normocin for Microbial contamination elimination before? Did it work?
I agree that we should do the microorganism identify tests to confirm what exactly we're facing. But we have no available equipment in the lab and we're in the rush to maintain the cells.
So I appreciate your valuable experience.
Does the medium get yellow(ish) and turbid? Bacteria usually are growing quite fast, when no (suitable) antibiotics are present.
I'd change the medium frequently and try as many antibiotics as I could get hold of.
Try to identify and eliminate the source of the contamination.
