I want to use LNCaP for some molecular studies.So,I just wanted to know the highest passage number of this cell line which can be used for these kind of studies
Very difficult to answer. In simpler terms, its advisable to cryopreserve cells very early in their passage and use early passage cells (probably less than 20-30 passages) to avoid genetic or epigenetic changes, which cancer cells frequently undergo due to dysregulation in growth control. I doubt any literature exists related to your question.
Normally, if you have tumor cell lines from collections, they have to be cultivated properly without any selection. Special rules about cell passages we use usually for primary cells in culture, so just be careful and look for the morphology of the cells to keep the same type as at the beginning.
I havent worked with this cell line but with other cancer cells, we usually dont go beyond passage 18. But I have a serious question for all the people who have commented. Are the cells actually in passage number 10 or 12 or 15 what you say.. Like some one said earlier that culturing the new cells and freezing them and using the freezed cells again .. But are these cells also in actual passage range.
Actually, passage number is actual in cell collections (depositories). We hope, that all people are correct with passage counting, but sometimes it is not. Anyway, it is very important for normal cells and primary cultures of normal cells. Most of cancer cells practically have no limits of passages.
