I'm currenctly working on the project CRISPR CAS9 transfection of U2OS cells with lipofectamine 3000.
The question I have: why after transfection different cel lines produce differen kind of clones, why for example the clones of iPS are more round and look like clogs and for U2OS it's just cells which are growing from one cell.
I'm really struggling to find more information about the formation of the clones from different kind of cells. Are there any suggestions where I can read more?
I've attached two foto's: one from iPS clones and another one from U2OS to make it clear.

U2-OS cells are more epithelial in structure (i.e. flat, broad) than stem cells. The colonies appear different simply because the cell types are quite different.
The clumpy nature of stem cell colonies is very characteristic of that cell type. In fact, if you were to uses U2-OS cells to generate induced pluripotent stem cells, you would see their morphology change from flat sheets to clumps (much like the fibroblasts vs the stem cells in the first picture you added).
