Hi All, I am learning to do antibody phage display, and really want to know what value for each steps can be considered as "normal" or "typical", so I can use as a reference when performing the bio-panning by myself. Greatly appreciated!

1. When producing helper phages, or phages contain phagemid, what yield (in pfu/mL or cfu/mL) is considered "normal"? For example, if I culture the E.coli under 30oC overnight.

2. When doing phage-ELISA, using phages expressing ScFv to bind with coated antigen, what EC50/Kd is considered "acceptable" or "normal"? I am familiar with antibody or ScFv-based ELISA to test the binding of Antibody to antigen, normally less than 10ng/mL for an antibody is considered "good" for me, but I just have no idea what value can be considered "normal" if I am using a phage rather than antibody for such ELISA.
