I am planning to design an in vivo dosing scheme for Paclitaxel (HY-B0015, MCE). Your technical support recommends using a mixed solvent of 10% DMSO + 40% PEG300 + 5% Tween-80 + 45% saline to dissolve Paclitaxel.
How well is this solvent system tolerated in rodents (e.g., mice/rats)? Is there any literature or experimental data to support this?
Is it possible that the concentration of each component (especially the 10% concentration of DMSO) may induce animal toxicity? Is it necessary to adjust the ratio?
If the drug is administered intraperitoneally/intravenously, is it necessary to pay attention to the injection volume limit or dosing interval?
The laboratory found that the dissolution effect of this system was good in the preliminary experiment, but is concerned about the impact of the solvent on animal health.
