EdU (HY-118411) labeling principle and reaction mechanism

The EdU marker (working concentration is usually 10-100 μM) is introduced into the cell culture system. As a thymine analog, it can be taken up by S phase cells and incorporated into newly synthesized DNA chains.

The detection relies on click chemistry reaction:

through the Cu (I)-catalyzed azide-alkyne cycloaddition reaction (CuAAC), the acetylene group in the EdU molecule is covalently coupled with the fluorescent labeled azide (such as the Alexa Fluor series probe) to form a stable triazole ring structure, realizing the visualization of the fluorescent signal of proliferating cells.
