I have a powdered compound which is not readily soluble in serum containing medium. To dissolve properly, I have made a stock solution of 100 mg/ml in 1 ml PBS of that compound. If I want to make a 10 mg/ml sample in serum containing medium, is it okay to take 100 ul of stock solution and mix that one in 1 ml of serum containing medium? Is it okay to serial dilute when there is two different solvent: PBS and serum containing medium?
That is not a problem; just keep track of the amount of PBS you carry over into your serum samples; calculate the dilution factor for the serum.
However, are you sure that your compound dissolved in PBS does not crash out, once it comes into contact with serum?

As long as the content of the stock solution doesn't interfere with what you intend to do it is OK. Ideally diluant should be used both to generate the initial stock solution and of course for serial dilution.
