As we know that the expression of PD 1 is less in jurkat cell. To express PD1 jurkat cell need to be stimulated or activated. But there is a chance of activation induced apoptosis. How this activation induced apoptosis can be reduced so that only cancer cell mediated apoptosis is highlighted. IS there any step to be followed after stimulation of T cell so that activation induced apoptosis is reduced?

You may add melatonin at different concentration in the range of 0.125 – 1mM to anti-CD3–stimulated cells as melatonin is known to protect T cells from anti-CD3–induced apoptosis.
TCR/CD3 stimulation results in rapid CD95L upregulation, and CD95L-mediated CD95 oligomerization is believed to be the primary mechanism of activation-induced cell death (AICD). Melatonin prevents AICD prior to CD95 engagement indicating that melatonin blocks early events of TCR/CD3-induced apoptosis.
