I'm sure we all here know the basics of chromatography, but what are the nifty tips and tricks that you have encountered over the years to aid in your method development? What modifiers (e.g. triethylamine), buffers, ion-pair reagents, unconventional mobile phase combinations, or salts have been particularly effective in your darkest hours when nothing else worked?

Use the most 'modern' technology like smaller particle sizes, smaller columns, pure and particle free mobile phases. Remember, it is the molecular target (API) that determines the method of analysis (sample matrix is 2nd).
