I amplifly ITS2 DNA from old mosquito samples preserved on naphthalene using antioxidant, but the target sequence does not amplify, only nonspecific fragments, smaller and more stable. This DNA would be fragmented? Fragments of mitochondrial DNA does not amplify as well.
The DNA of insects preserved with naphthalene is definitely gets degraded but can be amplified with lot of maneuvering. The chances of amplification will depends on for how long the insect has been preserved on naphthalene. Try with more quantity of DNA
using a higher quantity of DNA (if available) will not help to obtain reliable results because also ballast fragments will be amplified and a posteriori cleaning will decrease the amount of DNA wanted.
Try ethanol-preserved material if available.
Thank you so much Dr Anil and Dr Claus. I tested increasing DNA and really not amplified. Such sample contains many inhibitors and an eventual purification has not reproductivity.
