I want to design a fusion of GFP or YFP to the C- terminus of my protein. I am doing it for the first time so please help me with some golden rules -

1. What amino acid composition and length is the golden standard?

2. Second, and maybe more challenging - I would like to design the linker in such a way that there will be restriction sites located before and after the GFP tag so that after cutting and ligation a stop codon would be restored after my protein and GFP cut out? Has anyone done something like that before? Another way - I am looking for a double digest so that after ligation it restores the frame stop codon.
