Does any one know the reason behind the non-absorbance during MTT ASSAY even though the assay was carried out on the healthy fibroblastic cell culture. Are there any ways by which we can check the viability of the fibroblast cells apart from the MTT assay that can be carried out on the fibroblast cell???

The easiest way to check viability in cultured cells is trypan blue dye exclusion.

What reagents are you using to solubilise the formazan crystals? After your reaction period, the formazan is insoluble and needs to be released into the medium. We used DMSO or acidified ethanol, and it used to work ok. Alternatively to using MTT, you can also try using Alamar Blue/Resazurin. It works similarly to MTT, and is non-destructive to the sample, which can be advantageous.
