Make aliquots, repeated freeze thaw will decrease transduction efficiency and store at -80 ..I will not recommend at -20.

Use bleach for discard during experiment.

BSL2 is minimum requirement.

Thawing should be in ice only.

N calculate MOI carefully

After your experiment clean hood with iodine n ethanol and sterilize with UV.

-80C for long term storage (years). 4C for short term (few days)

Please be very careful for the second part of the question. Make sure you check the biosafety rules in your own university and institute. Ask your supervisor and biosafety officer. Normally, you have to obtain a permit first and also needs to be trained in order to handle the lentivirus correctly and safely.

Thanks Sandeep and Xia for your reply. Have you guys come across any reports that mentions the storage at -20 and its impacts on transduction? Or like what are your thoughts on storing at -20?
