I was wondering if anyone has any experience or an idea of the length and the concentration that would be useful for the incubation of PBMCs with oxytocin?

Figure 1.
Peripheral blood mononuclear cell proliferative response assessed by flow cytometric cell cycle analysis. The proliferative response of PBMCs to PHA and PHA plus oxytocin was evaluated as follows. Utilizing flat bottomed microtiter plates of 96 microwells (Falcon, Oxnard, CA, USA), 1.5×105 cells/well were cultured in triplicate in 200 μl of RPMI-1640 supplemented with 10% of autologous serum, 2 nmol glutamine (Flow Laboratories, Irvine, UK), 10 μg gentamicin (GIBCO, Invitrogen Ltd, Paisley, UK) and 1 μg of PHA (PHA-M; Boehringer, Ingelheim, Germany) or 1 μg of PHA plus 1×10−10 M, 1×10−11 M or 1×10−12 M of oxytocin (Sigma Aldrich).
Proliferative response of peripheral blood mononuclear cell (PBMCs) to phytohemagglutinin (PHA) and PHA plus oxytocin with and without oxytocin antagonist. The percentage of PBMCs in the S phase of the cell cycle after 72 hours of incubation with/without PHA, PHA plus oxytocin at 1×10−10 M, 1×10−11 M, 1×10−12 M) with or without oxytocin antagonist at 1×10−9 M are shown. Peripheral Blood Mononuclear Cells (PBMCs) Oxytocin Expression is Positively Correlated with Social and Behavioral Functions. Researchers suggest that higher peripheral OXTR expression as detected in PBMCs may reflect differential brain expression and allow improved response to prescribed oxytocin and/or future oxytocin derivatives/agonists.
