Hello all; We are going to quantify the bacterial DNA of individual bacteria in feces at the GENUS level by PCR. Although we tried to find the primer sets from published papers, we were not able to find useful primer sets. Next, we tried to design primer sets using 16S rRNA sequences obtained from LPSN database (https://lpsn.dsmz.de/) and primer BLAST. However, we were not able to obtain specific primer sets of each bacterial genus. When we confirm the specificity of the primer sets by BLAST search, the results always included other bacteria or unknown sequences. Could you help us how can we find primer sets for the following bacterial genera?

Search for the genus specific published primers in literature first. These are common genus and most probably you can find the primers.
