We are facing a problem with growth curves reproducibility in our lab. Fresh cultures growing at exactly same conditions are showing a very different growing pattern,did some one already have this problem?
Not really sure what the reason might be, but I have a couple of questions/comments:
Do you have any variability in the medium you are using (i.e. were media produced at the same time, using the same batches of chemicals)?
Does your medium contain peptone? These microbes are known to be very sensitive to bile salts which are one of the components in some of the main brands (and their exact concentration might vary from batch to batch. Maybe this would be a factor.
Are you facing reproducibility issues with subsequent culturing only, or have you tried doing simultaneous duplicates or triplicates of your growth experiments that you also have issues with?
