I dissolved the drug in DMSO and its completely dissolved but when I diluted in cell culture media formed crystals. I used (2-hydroxypropyl)-beta-cyclodextrin for dissolving the same small molecule for VIVO study. Can I use (2-hydroxypropyl)-beta-cyclodextrin as a solvent for Vitro study also? if I can, up to which concentration is possible to use it?

not sure about your new solvent, but maybe consider the solubility of your small molecule in aqueous solution (which media mainly is). If your final concentration you want to use in your assay is above that solubility border a different solvent of the stock solution will not help.
Sometimes we dissolve our compounds in media and using this as the highest concentration instead adding them as a stock solution to the media.

I hope u know that Beta Cylodextrin is not a solvent, its a solubilizing agent. it will form complex with hydrophobic material and keeps it in solution.
I dont know whether anybody used it.
theoretically u can use this in cell culture, but u need to show (by taking minimum concentration, that needs to be worked out by you) that it is not interfering with your study.
for example if u find 5micro molar is giving you the result u wanted, the real effective concentration will be lot lesser than that (as the the molecules in solution and in complex are in dynamic equilibrium). while explaining your results you always need to always mention that it is in cyclodextrin, tell the mechanics and justify it.
