i am analysing data obtained from technical experiment of treated same count of cell lines with two different drugs and their combination. Then i need to find out whether there is a synergy for the combination! the issue is that i treated same cell number for either drug A, drug B, to combination. is it logic to go ahead to compare the sum effect of both drugs to combination without normalization?
I normally have a brief look at CDI (coefficient of drug interaction) ; if there is something promising, do CalcuSyn which gives me publishable data.
Because we are treating the combinitorial well with two drugs for the same cell count as single drugs which are added to other drugs but the point of either; double-drugs on equal cell count in combination compared to each single drug treatment, or the double-cell count that results from the obtaining of the sum effect of single drugs compared to a half of cell count of experimental combinatorial treatment? does it make sense, thank you?
One way to normalize is to refer the data obtained to IC50 (or Dm) with the TC Chou method:
fa/fu = (D/Dm)m
