In my experience I used (Sigma) TRIZMA and Glycine 7 years old and I perform WB for pAKT, pp38 and pp44/42 without problem. If it was, at least, well conserved I think that you can try to use the powder to run a gel test. In any case pay attention, if the solution became quickly yellow it is better buying new reagents.

I would not expect any problems, especially if you can titrate and it holds the pH as expected. You need it to buffer and if it buffers it buffers :)

Cant exclude problems of course but this seems likely enough to be ok, that I would give it a try with the reagent you found. Especially, if its stored dry and dark.

thanks! i indeed found that bio-rad link (after reading on the container that stated that the shelf life could be extended if kept cold, which evoked my suspicions) and did not feel like making a huge 10X stock from it. maybe i'll start in a smaller scale :)

Thanks for posting this question. Why dry/powder reagents can indeed last super long especially if they are stable (maybe simpler the structure the stabler?). Wow, 14 years in storage and who knows how long it was in the warehouse before being labelled yet it may work. Please post here if it worked fine! Best wishes!

Only the amine group is remotely reactive.  If while stored it did not come in contact with an aldehyde, then the shelf life would be a very very long time.  To test how much break down you have, titrate a standard strong acid verses a known (analytically measured) amount of Tris.  If the amine group has been destroyed so will the buffer capacity of Tris.
