Hi all,

I am currently in need of a way to propagate Sendai virus (Cantell strain ATCC VR-907). I am aware that the traditional method of propagating Sendai is by infecting embryonate chicken eggs and harvesting the allantoic fluid, but I would prefer to avoid this method if possible. I am curious if anyone has had success in propagating Sendai in cell culture.

I have tried to propagate in the following conditions with no success:

MDCK CRL-34 with 3ug/mL Trypsin/TPCK

MDCK.2 with 3ug/mL Trypsin/TPCK

Vero with 3ug/mL Trypsin/TPCK

Regardless of the time of incubation (out to 7 days) I have failed to detect CPE to any notable amount and the supernatants also fail to hemmaglutinate chicken RBCs (the stock virus has a very high HA titer).

I have seen some information on the addition of DEAE-dextran to improve infection but I have not tried this.

Does anyone know of any protocols or have any experience with propagating Sendai virus in cell culture?
