I want to prepare a monolayer of adherent cells grown on cover slips for SEM. I have previously only prepared gels and am wondering if I could use the same fixation/dehydration protocol for the cell monolayer. My current protocol consists of fixation with 4% Glutaraldehyde at 4°C over night, dehydration in EtOH series (30%,40%,50%,60%,70&,80%,90%,96%,100%) and then chemical drying with HDMS.

Yes, you can use your protocol. The only difference: I usually use 2.5% glut. for 1-1.5 hrs at room temperature.
