Has anybody attempted to isolate RNA from low numbers (~5,000) unstimulated CD8+ lymphocytes? I've attempted using the usual kits (Qiagen RNA/DNA micro; Qiagen RNA micro, Zymo Direct-zol Microprep) and am about to have a go with Miltenyi micro-MACS RNA extraction but they seem to be very transcriptionally quiet!
I have found I need at least 20,000 cells to get any RNA output (using Qubit 4) from the kit whereas they work fine with as few as 500 MEFs. I'm unlikely to be able to sort that number from my patient samples.
My downstream application is RNA-seq.
